A study was performed on the numbers and species diversity of thermophilic fungi (growing at 45 °C in vitro) in 38 nests of 9 species of wetland birds, taking into account the physicochemical properties of the nests and the bird species. It was found that in nests with the maximum weight (nests of Mute Swan), the number and diversity of thermophilic fungi were significantly greater than in other nests, with lower weight. The diversity of the thermophilic biota was positively correlated with the individual mass of bird and with the level of phosphorus in the nests. The dominant species within the mycobiota under study was Aspergillus fumigatus which inhabited 95 % of the nests under study, with average frequency of ca. 650 cfu g?1 of dry mass of the nest material. In a majority of the nests studied (nests of 7 bird species), the share of A. fumigatus exceeded 50 % of the total fungi growing at 45 °C. Significantly higher frequencies of the fungal species were characteristic of the nests of small and medium-sized piscivorous species, compared with the other bird species. The number of A. fumigatus increased with increase in the moisture level of the nests, whereas the frequency of occurrence of that opportunistic pathogen, opposite to the general frequency of thermophilic mycobiota, was negatively correlated with the level of phosphorus in the nest material, and with the body mass and length of the birds. The authors indicate the causes of varied growth of thermophilic fungi in nests of wetland birds and, in particular, present a discussion of the causes of accumulation of A. fumigatus, the related threats to the birds, and its role as a source of transmission in the epidemiological chain of aspergillosis.  相似文献

One hunderd and ninety five wild pigs from two different regions of Poland were investigated for transferrin, amylase and ceruloplasmin polymorphism. A new transferrin phenotype Tf PB was detected. This phenotype differed from Tf AB in the electrophoretic mobility of the more anodal transferrin. Tf P is assumed to be the product of a new allele Tf P at the Tf locus. Two amylase phenotypes Am 1–2 and Am 2 were observed. The Am 1 allele was absent from the pigs in the Poznan region. Only one ceruloplasmin phenotype, Cp B, was found.  相似文献

We investigated temporal changes in diet composition of the Montagu''s Harrier Circus pygargus breeding in natural habitat (calcareous peat bog) in SE Poland. We characterized diet composition in a three‐year period (2007–2009), based on pellet analyses. We investigated whether diet composition was affected by years or stage of breeding. We compared diet of the studied population between 2000s and 1990s and with other populations. We found that the food of the studied population was dominated by insects and mammals (by number) and mammals and birds (by biomass). Biomass and abundance of main prey items differed between studied years because of different air temperatures. We found some interannual differences in contribution of some prey items including higher number of thermophilic prey (insects and amphibians) in warmer years. Comparison of pellet composition in the 1990s and 2000s revealed significant increase in the abundance of thermophilic prey (insects and reptiles) and decrease of mammals including Microtus voles and birds. Those changes may be linked to habitat changes in areas neighboring peat bogs and climate change‐induced changes in prey communities. The studied population was able to respond to changes in foraging habitats and prey composition by opportunistic foraging on easily available prey. The diet of the studied population is the most similar to the geographically closest populations foraging in similar habitats and characterized by high contribution of insects.  相似文献

We used time-dependent fluorescence energy transfer, time-dependent collisional quenching, and global analysis of the data resulting from these through-space and contact interactions to recover the end-to-end distance distributions and diffusion coefficients of flexible fluorescent molecules. The fluorescence decays of covalently linked tryptamine-acceptor and tryptamine-quencher pairs were measured by the frequency-domain method. These data were fit using numerical solutions of the differential equation, which predicts the time- and distance-dependent population of the excited state donors in the presence of energy transfer or collisional quenching, followed by transformation to the frequency domain for nonlinear least-squares comparison with the experimental data. We found that the energy transfer data for the donor-acceptor pair alone were adequate to recover the starting distribution and the end-to-end diffusion coefficient; however, the resolution is dramatically improved by the use of both the through-space and contact interactions.  相似文献

Quercetin is a naturally occurring flavonoid that has a lot of beneficial properties to human health. In this report, using the spin label technique, the influence of quercetin on the fluidity of multilamellar DPPC liposomes was studied. The polarity of the environment preferred by quercetin was also examined by determining the dependence of the position of electronic absorption maxima on dielectric properties of different environments. Autofluorescence of quercetin was also used to examine its distribution in cells. An additional aim of the study was to find how quercetin presence affects human skin fibroblasts. The results showed that incorporation of quercetin at physiological pH into DPPC liposomes caused changes in the partition coefficient of the Tempo spin label between water and polar head group phases. By determining the electronic absorption maxima, we observed that the chromophore of quercetin is localized in the polar head region. Fluorescence microscopy of HSF cells showed quercetin presence in the membrane, cytoplasm and inside the nucleus. Ultrastructural observation revealed some changes, especially in membranous structures, after flavonol treatment. From the results we have concluded that quercetin present in the membrane and other structures can cause changes within cells crucial for its pharmacological activity.  相似文献

Cyt1A is a cytolytic toxin from Bacillus thuringiensis var. israelensis. A computer model of the toxin in solution was generated and validated by resonance energy transfer (RET). The average distance between the two tryptophans (residues 158 and 161) and the fluorescently labeled cysteine 190 was 2.16 nm, which closely matched the distance predicted in computer simulations, 2.2 nm. The simulation results were able to explain two previous experimental observations: (i) amino-acid sequences of all Cyt toxins contain four blocks of highly conserved residues; and (ii) several single-point mutations drastically abrogated Cyt1A's toxicity. Selective randomization of atomic coordinates in the computer model revealed that the conserved blocks are important for proper folding and stability of the toxin molecule. Replacing lysine 225 with alanine, a mutation that renders the toxin inactive, was shown to result in breaking the hydrogen bonds between K225 and V126, L123, and Y189. Calculated Helmholtz free energy difference of the inactive mutation K225A was higher by 12 kcal/mol and 5 kcal/mol than the values for the benign mutations K118A and K198A, respectively, which indicates that the K225A mutant is significantly destabilized. The normal-mode and principal-component analyses revealed that in the wild-type Cyt1A the region around the residue K225 is quite stationary, due to the hydrogen-bond network around K225. In contrast, pronounced twisting and stretching were observed in the mutant K225A, and the region around the residue K225 becomes unstable. Our results indicate that conformational differences in this mutant spread far away from the site of the mutation, suggesting that the mutant is inactivated due to an overall change in conformation and diminished stability rather than due to a localized alteration of a “binding” or “active” site.  相似文献

We measured the frequency response of the polarized emission of Yt-base in propylene glycol at 10 ° C. Data were obtained for excitation wavelengths of 290, 312 and 346 nm, for which the fundamental anisotropies are 0.05, 0.19 and 0.32, respectively. Additionally, data were obtained using CCl4, to decrease the mean decay time from 9.1 to 4.2 ns. These nine sets of data were analyzed globally to recover the anisotropy decay law. Three correlation times were needed to fit the data, 0.8, 3.0 and 5.6 ns, a range of only 7-fold. We believe this is the first reported detection of three correlation times for a rigid molecule.  相似文献

Summary In this paper the literature data on the structure-activity relationship for the series of tuftsin analogs are summarized. Among others, the questions of the substitution of particular amino acid residues in different positions of the peptide chain, as well as the questions of shortening and lengthening of the peptide chain of tuftsin, are reviewed.
